Product description
Medicago’s SSC buffer is specifically developed for Northern and
Southern transfer protocols and nucleic acid preparations. SSC
buffer is an acid salt buffer solution and is used as a buffering
agent to control acidity. The buffer is supplied in two sizes giving
100 ml or 1000 ml of 0.30 M sodium citrate, 3.0 M sodium
chloride buffer or 1000 ml of 0.03 M Sodium citrate, 0.30 M
sodium chloride buffer with pH 7.0 at 25°C when one tablet or
the contents of one pouch is dissolved in deionized water.

• Nucleic acid hybridization
• Blot transfer procedures
• Nucleic acid preparation

Directions for use
Empty one pouch or deposit one tablet of the SSC buffer in a
laboratory flask or beaker placed on a magnetic stirrer. Add
50 ml or 300 ml of deionized water and stir for a couple of
minutes. Adjust the volume up to 100 ml or 1000 ml, stir until
full dissolution and the buffer is ready to use.

Tips and hints
Dilute the SSC buffer stock solutions to required concentration
just before use. If the tablet or the contents of the pouch is not
properly dissolved, make sure:
– the water temperature is 25°C (do not exceed this temperature)
– the buffer solution is properly stirred.
Sterilization can be performed by filtration. Filter the buffer
solution into a sterile flask through a 0.22 µm filter and keep
at +4°C.

Shipping and storage
SSC buffer is shipped at room temperature. Store the tablets
and pouches in a dry place at room temperature. Shelf life is
three years.
